Testing a flexible gene editing method called Free-BE in moss, maize, and wheat

Updated

Abstract

Free-BE achieved fewer indels and a higher proportion of single-nucleotide edits compared to the conventional fused system.

  • Free-BE demonstrated lower editing efficiency compared to the conventional Fused-BE system in the moss Physcomitrium patens.
  • The modular Free-BE strategy resulted in fewer undesired editing outcomes, such as bystander edits and out-of-window modifications.
  • Free-BE altered the spectrum of base substitutions, producing a higher proportion of C-to-G and C-to-A transversions.
  • No mutations were detected at four predicted off-target loci in P. patens.
  • In a maize protoplast reporter assay, Free-BE successfully achieved targeted base conversions.
  • Stable transformation of wheat showed successful editing of endogenous loci, although with lower efficiency than Fused-BE.
