A new mouse model for a high-risk age-related macular degeneration gene variant, ARMS2 A69S

Updated

Abstract

By 12 months of age, ARMS2 A69S mice developed AMD-relevant phenotypes.

  • The highest genetic risk for age-related macular degeneration (AMD) is associated with the region on Chromosome 10q26.
  • Functional studies of the gene ARMS2 have been limited due to the absence of animal models in rodents.
  • The complete human ARMS2 gene carrying the A69S risk variant was successfully inserted into a mouse model.
  • RNA analysis showed specific patterns of ARMS2 expression, particularly in neural tissues and the eye.
  • Mice with the ARMS2 A69S variant exhibited increased fundus spots and thinning of the outer nuclear layer.
  • Transcriptomic profiling indicated reduced mitochondrial oxidative-phosphorylation genes and lower retinal mtDNA copy number.
