High-Resolution MRI Tracking of Early Fatty Liver Disease and Treatment Response in a Mouse Model Before Scarring

Abstract

Histology confirmed early metabolic dysfunction-associated steatohepatitis (MASH) after 16 weeks of a high-fat and high-cholesterol diet, without fibrosis.

  • Multiparametric MRI parameters showed strong correlations with histopathological scores.
  • Proton density fat fraction (PDFF), liver fat content (LFC), and T-values increased significantly by 6.8-fold, 5.2-fold, and 2.5-fold, respectively, while the apparent diffusion coefficient (ADC) decreased by 30% (p < 0.001).
  • Semaglutide treatment led to a 73% decrease in PDFF, 62% reduction in LFC, and 46% decrease in T-values (p < 0.001), while ADC increased by 1.4-fold (p = 0.017) compared to the vehicle group.
  • The established MRI evaluation system allows for noninvasive, longitudinal monitoring of early MASH dynamics and treatment response.
