mRNA therapeutics delivered by lipid nanoparticles (LNPs) have the potential to address neurodegenerative diseases but have not yet been tested in clinical trials.
- Recent advances in engineering LNPs may enable them to cross the blood-brain barrier and selectively express in ageing brain cells.
- Emerging chemistries, such as BBB-shuttling lipids and peptide-functionalized shells, could facilitate systemic delivery to the central nervous system in animal models.
- First-in-human studies for rare metabolic disorders indicate favourable safety profiles and manufacturability for mRNA-LNPs.
- Challenges remain in achieving efficient delivery and expression durability in aged neurons and addressing chronic-dose immunogenicity.
