mTREX2 increases large DNA deletions during CRISPR editing of complex sugarcane genome

Updated

Abstract

Editing efficiencies ranged from 31% to 85% in sugarcane using the LbCas12a system.

  • The LbCas12ai-RRV variant consistently demonstrated higher overall and allelic editing efficiencies compared to LbCas12ai-R.
  • Co-expression of the mouse repair enzyme mTREX2 enhanced the frequency of larger deletions in the editing process.
  • The frequency of deletions of 51 base pairs or more increased approximately 7-fold with both editing variants.
  • Targeted mutagenesis of the lw1 gene led to observable changes in sugarcane, affecting chlorophyll and carotenoid production.
  • Capillary electrophoresis-based genotyping offered a cost-effective and high-throughput method for assessing editing outcomes.
