Sticky coated lipid nanoparticles for oral delivery of docetaxel and curcumin: formulation, lab testing, and cancer cell toxicity

Updated

Abstract

TMC-coated solid lipid nanoparticles (SLNs) achieved a mean particle size of 136.7 ± 6.4 nm and an encapsulation efficiency of over 97% for both docetaxel and curcumin.

  • The optimized SLNs exhibited a spherical shape and a polydispersity index of 0.315 ± 0.037.
  • Gastric burst release was reduced to less than 2%, attributed to the protonation of TMC at acidic pH, enhancing gastrointestinal stability.
  • The drug release kinetics followed the Korsmeyer-Peppas model, indicating a non-Fickian diffusion mechanism influenced by polymer relaxation.
  • A cumulative release of 50.4% of docetaxel and 58.6% of curcumin was observed over 96 hours, with sustained and pH-triggered release.
  • In vitro cytotoxicity studies showed increased effectiveness of TMC-SLNs against MCF-7 breast cancer cells compared to free drugs and blank SLNs.
  • No toxic effects were noted in normal MCF-10 A cells, suggesting a favorable safety profile for the developed formulation.
