Lipid nanoparticle-encapsulated IFNα2 gene therapy suppresses colon cancer lung metastasis in mouse models.

  • The therapy selectively transfects tumor cells, restoring local IFNα2 production in lung metastases.
  • Efficacy of the treatment requires specific signaling through the IFN receptor (IFNAR1).
  • Neutralizing co-induced IL6 enhances the therapeutic effect.
  • Single-cell RNA sequencing revealed significant changes in the tumor microenvironment, including macrophage apoptosis and monocyte transformation.
  • Tumor cells exhibited increased antigen presentation and immune-cell-recruiting chemokines while shifting away from a program resistant to a specific form of cell death.
  • The treated tumor microenvironment mirrored the immune signatures found in patients who responded to pembrolizumab therapy.
