Effectiveness and safety of the drug KarXT for schizophrenia in the USA: results from a phase 3 clinical trial

Updated

Abstract

The trial met its primary endpoint with a mean change in PANSS total score of -21.2 points for KarXT compared to -11.6 points for placebo.

  • KarXT demonstrated significant improvement in symptoms of schizophrenia compared to placebo, with a least squares mean difference of -9.6 points.
  • All secondary endpoints were also favorable for KarXT, achieving statistical significance (p<0.05).
  • Common adverse events associated with KarXT included constipation (21%), dyspepsia (19%), and nausea (19%), with rates generally higher than placebo.
  • No extrapyramidal motor symptoms were reported in the KarXT group, indicating a potential for lower risk of these side effects compared to traditional antipsychotics.
  • The results suggest that KarXT may represent a new class of antipsychotic medications that activate muscarinic receptors rather than blocking dopamine receptors.
