Muscle loss linked to drugs that activate the glucagon-like peptide-1 receptor: A study of population data

Abstract

A total of 142 cases of muscle atrophy were identified with glucagon-like peptide-1 receptor agonist therapy.

  • Muscle atrophy cases were predominantly reported in adults aged 18-64 years, with 43% female and 57% male.
  • Semaglutide showed a reporting odds ratio (ROR) of 2.39, indicating increased reporting of muscle atrophy compared to all other drugs.
  • Tirzepatide also showed a significant ROR of 1.69 for muscle atrophy, suggesting higher reporting rates.
  • In contrast, exenatide and liraglutide were associated with significantly lower reporting odds of muscle atrophy, with RORs of 0.26 and 0.27, respectively.
  • These findings point to a need for further studies to objectively measure muscle mass and function in relation to GLP-1 RA therapy.
