Nanocarrier-delivered simvastatin may affect energy regulation and oxidative stress in an Alzheimer's disease model

Abstract

Simvastatin delivered via a nanocarrier showed a substantial reduction of Aβ plaques in a rat model of Alzheimer's disease.

  • Simvastatin and its nanocarrier enhanced the process of cellular recycling by increasing specific protein levels associated with autophagy.
  • Both forms of simvastatin demonstrated strong antioxidant and anti-inflammatory properties by decreasing markers of oxidative stress and inflammation.
  • Administration of simvastatin increased levels of acetylcholine and lowered the expression of the acetylcholinesterase gene.
  • Histopathological analysis indicated reduced neuronal degeneration and glial activation with near-normal brain structure in the nanocarrier group.
  • The results suggest that simvastatin may provide neuroprotective effects through multiple biological pathways in Alzheimer's disease.
