Abstract

Out of 5,425 candidate neuronal enhancers tested, 2,422 were found to be active in human neurons.

  • Massively Parallel Reporter Assays identified active enhancers that could be targeted for gene activation.
  • Multiplex single-cell CRISPRa screening assessed 15,643 guide RNAs across enhancer and promoter regions.
  • Hundreds of CRISPRa guide RNAs were found to upregulate 200 out of 337 neurodevelopmental disorder genes.
  • Ninety-one novel enhancer-gene pairs were identified that may play a role in neurodevelopment.
  • Certain CRISPRa guide RNAs showed selective upregulation of target genes when tested in various human and mouse models.
