Gene Editing Treatment Nexiguran Ziclumeran for Hereditary ATTR with Nerve Damage

Abstract

Essence

A single dose of nexiguran ziclumeran was linked to durable transthyretin lowering and generally stable neuropathy measures in hereditary ATTR polyneuropathy.

Evidence

Phase 1 open-label study in 36 patients with ATTRv-PN found mean serum TTR reductions of 90% at day 28 and 92% at month 24, with mean mNIS+7 change of -8.5 over 27 months of follow-up.

Caveat

This was a small uncontrolled phase 1 trial, and serious adverse events occurred in 11 patients, including 1 death from cardiac amyloidosis and 1 withdrawal for motor decline.
