Rapid visual test using omp25 and CRISPR technology to detect Brucella DNA linked to brucellosis

Updated

Abstract

The visual recombinase polymerase amplification (RPA)-CRISPR/Cas12a assay can detect as low as 2.72 copies/μL of Brucella DNA.

  • The assay targets a 642-bp region of the Brucella omp25 gene.
  • Fluorescence detection occurs within 20 minutes for both RPA amplification and Cas12a reaction.
  • No cross-reactivity was observed with non-target pathogens during testing.
  • Among 96 bovine nasal swab samples, the assay demonstrated a positive percent agreement of 100.00% with qPCR results.
  • The assay is not intended for differentiating between vaccine-derived and wild-type Brucella DNA.
