Risk of Optic Nerve Damage with Incretin-Based Diabetes Treatments: A Comparison Using Real-World Safety Data

Updated

Abstract

Semaglutide has 355 reported cases associated with optic ischaemic neuropathy, indicating a strong post-marketing safety signal.

  • Tirzepatide and liraglutide also showed significant safety signals with 49 and 13 reports, respectively.
  • The reporting odds ratio for semaglutide was 94.45, suggesting a markedly higher risk compared to other treatments.
  • No safety signals were found for dulaglutide, exenatide, or lixisenatide, indicating potential differences within the class.
  • Among the reports for semaglutide, the mean age of patients was 59 years, with 18% experiencing disability and 11% requiring hospitalization.
  • The findings suggest that the safety profiles of these incretin-based therapies may not be uniform.
