Improving Oral Semaglutide Tablets to Boost Stomach and Intestine Absorption Using Sodium Caprate

Sodium caprate (C10) enhanced semaglutide absorption to levels comparable to SNAC-based formulations.

  • C10 demonstrated sufficient buffering capacity to neutralize acidic environments, aiding absorption.
  • In cell and animal studies, C10 increased semaglutide absorption in a concentration-dependent manner.
  • The optimized monolayer tablet formulation showed the highest similarity in dissolution to Rybelsus.
  • Pharmacokinetic parameters from the monolayer formulation in beagle dogs overlapped with those of the SNAC-based product at matched doses.
  • These findings suggest that C10 could serve as a viable alternative to SNAC in oral peptide delivery systems.
