Early study of increasing doses of orforglipron in Japanese people with type 2 diabetes

Essence

Orforglipron was tolerated in this small Japanese type 2 diabetes phase 1 study and showed reductions in glucose, HbA1c, and body weight over 12 weeks.

Evidence

This double-blind randomized placebo-controlled phase 1 single- and multiple-ascending-dose study enrolled 23 participants in Part A and 60 in Part B, with daily oral orforglipron up to 45 mg or placebo for 12 weeks.

Caveat

The trial was small, short, and phase 1, so efficacy and safety need confirmation in larger longer studies.
