CRISPR tests and humanized liver mice identify host factors for hepatitis B virus

Abstract

A pooled, genome-wide CRISPR-Cas9 knockout screen identified 72 genes associated with Hepatitis B virus (HBV) infection.

  • Depletion of the antiviral factor EXOC1 enhanced multiple HBV replication markers.
  • Transcriptomic analysis indicated activation of hypoxia- and HIF-1-related gene signatures following EXOC1 depletion.
  • In vivo experiments with a human liver chimeric mouse model showed that knockout of the proviral factor ZCCHC14 significantly reduced HBV replication and spread.
  • The study integrates findings from both in vitro and in vivo experiments to expand the understanding of HBV host factors.
