PEG-lipid composition significantly influences the structural and functional properties of siRNA lipid nanoparticles (LNPs).

  • Colloidal properties such as size (<120 nm), polydispersity index (<0.2), and encapsulation efficiency (≥ 90%) are maintained across different LNP formulations.
  • The presence of mixed PEG-lipids leads to a lower apparent pKa and a broader transition upon pH change, indicating a more complex interfacial environment.
  • LNPs show medium-dependent structural and surface changes due to PEG shedding and protein interactions.
  • Formulations with mixed PEG-lipids exhibit intermediate shedding kinetics and improved adaptability of the surface.
  • Variations in PEG-lipid composition result in differences in cellular uptake and gene silencing kinetics in HCT116 cells.
