Abstract
A novel PET probe, [18F]AlF-NOTA-Asp2-tirzepatide, was developed to visualize tirzepatide's in vivo distribution.
- The probe primarily eliminated through the renal system.
- Elevated uptake in the heart, liver, and blood was observed due to enhanced albumin binding from a C20 fatty diacid moiety.
- In vitro assays indicated reliable binding affinity for GLP-1R and moderate binding to GIPR.
- Persistent tumor accumulation was noted in INS-1 and A549-GIPR xenografts, enabling effective visualization of tumors.
- Non-target uptake remains excessive and requires optimization for potential clinical use.
