How plasmid design affects the stability of end DNA repeats in adeno-associated virus vectors

Updated

Abstract

An analysis of 7,041 AAV plasmids revealed a widely disseminated 11-bp ITR deletion in 4,773 plasmids.

  • ITRs were highly stable during short-term culture, but prolonged culture showed positional effects leading to preferential loss or mutation of the nearest ITR.
  • Among two-ITR plasmids, 95.3% of the deletions were found in the origin-proximal ITR.
  • A novel rAAV entry vector with stable full-length native AAV2 ITRs was constructed for efficient packaging of a 4,750-bp CRISPR-Cas9 cassette.
  • Preliminary evidence suggests that ITR sequence variation may influence genome integration outcomes.
