Polo-like kinase 1 as a treatment target in aggressive neuroblastoma

Abstract

PLK1 is highly expressed in unfavorable neuroblastoma and is associated with adverse patient outcomes (P < 0.001).

  • Expression of PLK1 correlates with unfavorable prognostic markers, including stage 4 and age greater than 18 months.
  • Inhibition of PLK1 with nanomolar doses of BI 2536 leads to significantly reduced proliferation in all analyzed neuroblastoma cell lines.
  • Cell treatment with BI 2536 results in cell cycle arrest and increased cell death.
  • BI 2536 effectively halts the growth of neuroblastoma xenografts in nude mice.
