Adding multiple adenine tails without a template boosts CRISPR/Cas12a activation for stronger lateral flow biosensors

Updated

Abstract

The assay enables sensitive detection of TdT with a limit of detection of 0.016 U/mL.

  • A lateral flow assay converts TdT activity into a visual signal using CRISPR/Cas12a technology.
  • TdT catalyzes poly-adenine extension, which enhances the activation of Cas12a for signal amplification.
  • The assay shows high specificity for TdT compared to other polymerases.
  • Performance in human serum samples resulted in recoveries between 98.8% and 103.7%.
