Abstract

Post-loaded lipid nanoparticles (LNPs) maintain mRNA delivery efficiencies comparable to conventional LNPs.

  • Post-loaded LNPs were created by first fabricating empty LNPs and then loading RNA, rather than mixing lipids and RNA simultaneously.
  • Colloidally stable empty LNPs were produced by optimizing pH, buffer composition, lipid concentration, and ethanol content.
  • Loaded LNP diameters were maintained below 100 nm while incorporating four distinct RNA payloads efficiently.
  • Post-loaded LNPs showed similar internal architectures and surface properties to conventionally co-precipitated LNPs.
  • This approach allows for decentralized assembly of mRNA LNPs, enabling storage under mild refrigeration.
