Targeting polycomb repressive complex 2 (PRC2) enhances the antitumor activity of 19BBζ CAR T cells.

  • Inhibition of PRC2 consistently led to the development of granzyme B+ effector memory CD8 T cells.
  • PRC2 inhibition promoted the long-term persistence of granzyme B+ effector memory CAR T cells.
  • CAR T cells with PRC2 inhibition demonstrated sustained antitumor activity in both lab and animal studies.
  • PRC2-inhibited CAR T cells did not show signs of exhaustion over time.
  • Differentiation of patient-derived anti-CD19 CAR T cells was enhanced through TCR restimulation combined with PRC2 inhibition.
