Preventive and treatment effects of mRNA vaccines targeting HPV16 proteins in early-stage HPV tumor mouse models

Abstract

E7 mRNA/lipid nanoparticle vaccination led to significant tumor regression and undetectable HPV16 E6/E7 DNA in a murine model.

  • E7 mRNA/LNP induced stronger T cell responses compared to E6 in a low-dose setting.
  • In therapeutic settings initiated shortly after tumor implantation, E7 vaccination resulted in robust T cell responses and significant tumor regression.
  • A two-dose regimen of E7 vaccination prevented tumor establishment after TC-1 cell inoculation.
  • E7 vaccination demonstrated durable immune memory, with sustained tumor protection and undetectable HPV DNA upon rechallenge at 5 months.
  • These findings support the potential of E7 mRNA/LNP as a therapeutic vaccine for HPV-associated diseases.
