Controlling the DNA-cutting activity of LbCas12a by adjusting temperature

Updated

Abstract

A limit of detection (LOD) of 0.73 aM was achieved against Mycobacterium tuberculosis using a novel thermal-regulated CRISPR-Cas12a assay.

  • Temperature-regulation using cost-effective ssDNA blockers allows precise control of Cas12a activity.
  • The assay operates effectively within a narrow temperature range of 5°C (37-42°C).
  • Different DNA blockers can activate the cis- and trans-cleavage activity of Cas12a at desired temperatures.
  • The one-tube thermal-start RPA-CRISPR/Cas12a assay shows 96.67% concordance with qPCR in clinical specimens.
  • A 3D-printed portable device was developed for streamlined clinical testing, demonstrating 90.48% sensitivity and 100% specificity.
