Using blood proteins to predict risks of age-related diseases

Abstract

A total of 185 PhenoAge-related plasma proteins were utilized to develop a new aging clock called ProtPhenoAge.

  • ProtPhenoAge demonstrated a strong correlation with biological age (r = 0.96, R = 0.92).
  • The model showed a predictive capability for all-cause mortality with an area under the curve (AUC) of 0.76.
  • ProtPhenoAgeAccel was significantly linked to 313 disease phenotypes related to aging.
  • Compared to previous aging clocks, ProtPhenoAge identified a greater number of age-independent but aging-related phenotypes.
  • In a genome-wide association study, 10 aging-associated genetic regions were identified, including loci related to epigenetic aging and the APOE gene.
