Adenine Base Editing Fixes Cell Defects Caused by PRPF8 Mutation in Human Retinal Cells Made from Stem Cells

Abstract

PRPF8-mutant retinal pigment epithelium (RPE) cells showed decreased PRPF8 mRNA and protein levels.

  • Mutant RPE cells displayed weakened barrier function and disrupted cell polarity.
  • Loss of apical microvilli and pigment granules was observed in ultrastructural analysis.
  • Transcriptomic analysis revealed abnormal splicing events, particularly in cilium assembly and melanosome pathways.
  • Correction of the PRPF8 mutation using adenine base editing restored PRPF8 expression.
  • Restoration of normal barrier integrity and apicobasal polarity was achieved after gene correction.
