Psilocybin activates the stress system differently depending on sex and situation

Psilocybin may strongly activate the HPA axis via specific neurons in the hypothalamus.

  • Psilocybin could enhance the activity of corticotropin-releasing hormone neurons in the hypothalamus, particularly in female mice.
  • Activation relies on serotonergic 5-HT2A and 5-HT2C receptors.
  • There are two mechanisms for 5-HT2A receptor effects: direct activation of neurons and increased glutamate release.
  • Psilocybin may reduce neuronal activity in response to environmental changes, unlike typical stress responses.
  • This modulation could help explain how psychedelics may recalibrate unhealthy stress reactions.
  • Findings highlight the importance of context, or 'setting', in the effects of psychedelics.
