Blocking Rad51 protein helps stop DNA repair in glioma tumors and tumor stem cells

Abstract

Rad51 levels are upregulated in high grade gliomas compared to normal human astrocytes.

  • High grade gliomas show increased levels of Rad51, a protein involved in DNA repair.
  • Rad51 foci formation in established glioma cell lines increases with radiation and temozolomide treatment.
  • Higher Rad51 levels are associated with reduced sensitivity to radiation therapy.
  • Downregulation of Rad51 significantly enhances the cytotoxic effects of temozolomide.
  • Residual γ-H2AX foci persist longer after treatment with combined therapies when Rad51 is knocked down.
  • Rad51 levels in clinical glioma samples do not correlate with tumor grade.
