Fast Visual Test Combining DNA Amplification and CRISPR to Detect Pear Canker Pathogens

Updated

Abstract

The limit of detection for the developed system is 1 copy/μL of the target template.

  • A hybrid LAMP-CRISPR/Cas12a fluorescence detection system was created to identify Cytospora pyri and C. mali, the main pathogens causing pear canker.
  • The LAMP module amplifies the target within 30 minutes at 65 °C, followed by the CRISPR/Cas12a module producing stable fluorescence in approximately 15 minutes at 37 °C.
  • The system effectively detects both C. pyri and C. mali without cross-reactivity to other Cytospora species or common fruit tree pathogens.
  • Validation with diverse Cytospora strains showed complete agreement between LAMP, CRISPR/Cas12a fluorescence detection, and UV-assisted visual evaluation.
  • Preliminary results from Korla fragrant pear samples aligned with conventional PCR findings, indicating potential for use in orchard settings.
