Stress disrupts daily hormone patterns and harms egg development in mice

Abstract

Restraint stress caused sustained serum corticosterone elevation and increased glucocorticoid receptor expression in cumulus cells.

  • Corticosterone levels declined while ovarian gene expression of Cyp19a1, Esr2, and Bmal1 increased during late antral follicle growth.
  • Restraint stress suppressed the expression of Cyp19a1, Esr2, and Pkm2, disrupting expected temporal patterns.
  • During oocyte maturation, increased ATP demand and glycolytic gene expression were observed alongside decreased BMAL1 expression in cumulus cells.
  • Restraint stress reduced the proliferative capacity of cumulus cells and impaired the formation of tight junction plaques.
  • E2 supplementation improved the expression of Per2 and Bmal1 in cumulus cells and enhanced glycolysis, while Bmal1 knockdown negatively affected metabolic support.
