Drug treatments for adults with overweight and obesity: a combined analysis of clinical trials

A total of 49,810 participants were enrolled across 143 trials evaluating weight-lowering drugs for adults with overweight and obesity.

  • Phentermine-topiramate was identified as the most effective drug for weight reduction, with a significant odds ratio of 8.02 for achieving a weight loss of 5% or more.
  • GLP-1 receptor agonists also demonstrated substantial weight loss benefits, with an odds ratio of 6.33 for similar weight loss outcomes.
  • Naltrexone-bupropion and orlistat were found to be associated with increased adverse events leading to discontinuation of treatment.
  • Semaglutide, a GLP-1 receptor agonist, showed greater weight loss benefits compared to other drugs while having a similar risk of adverse events.
  • High to moderate certainty evidence supports the use of these pharmacotherapies for adults who struggle with overweight and obesity.
