rHuPH20 improves delivery of mRNA nanoparticles outside blood vessels in mice and minipigs

Abstract

Co-administration of recombinant human hyaluronidase PH20 (rHuPH20) increased local expression of mRNA lipid nanoparticles (mRNA-LNPs) by up to 38-fold in mouse studies.

  • In mice, rHuPH20 enhanced local expression of mRNA-LNPs by up to 7-fold after intramuscular administration.
  • Systemic exposure of mRNA-LNPs was increased by up to 38-fold following subcutaneous dosing when rHuPH20 was used.
  • In minipigs, subcutaneous administration of huEPO mRNA-LNPs with rHuPH20 achieved serum levels approaching those of intravenous administration.
  • rHuPH20 also improved hepatic expression of mRNA-LNPs after extravascular delivery.
  • The use of rHuPH20 was associated with reduced pro-inflammatory cytokine responses during repeat dosing.
