Using RNA helpers with CRISPR to create and isolate plants without Cas9 protein

Updated

Abstract

Essence

An RNA aptamer-based CRISPR/Cas9 reporter system improved edited-plant selection and faster recovery of Cas9-free mutants compared with GFP/Cas9.

Evidence

Plant genome-editing platform comparison showing higher T1 mutation rates, 30.2% better T2 sorting efficiency, and more efficient homozygous double-target mutant generation with 3WJ-4 × Bro/Cas9 than GFP/Cas9.

Caveat

This was a plant engineering system comparison, so the results show technical performance in that setting rather than broad effectiveness beyond the tested model.
