Rapid CRISPR Test for ALV-J Virus with Wide Detection and Lower Leftover DNA

Updated

Abstract

The one-tube RPA-CRISPR/Cas12a assay shows a lowest tested concentration yielding 9/9 positive reactions at 1 × 100 copies/μL.

  • A mixed crRNA-based CRISPR/Cas12a detection system may effectively cover most circulating ALV-J variant strains.
  • Optimizations in RPA mix and buffer conditions led to a simplified one-pot assay that consistently detects as low as 1 × 102 copies/μL.
  • For lateral flow assays, the detection limit was maintained at 1 × 101 copies/μL by reducing RPA amplification and using enhanced Cas12a protein.
  • Specificity tests showed no cross-reactivity with non-target avian viruses, confirming a positive signal only for ALV-J.
  • When tested on clinical samples, the one-tube assay matched RT-qPCR results in positive detection rates.
