The platform achieved a detection limit of 6.08 copies/μL and a linear quantitative range of 10-10 copies/μL.

  • A multi-unit microfluidic system allows concurrent construction of standard curves and sample analysis.
  • Signal generation is accomplished through a one-pot RPA-CRISPR/Cas12a assay with optimized amplification and cleavage kinetics.
  • Performance of the system is comparable to qPCR, with an area under the curve (AUC) of 0.984.
  • Intra- and inter-assay variability is acceptable, with relative standard deviations of 2.63-6.07%.
  • The method demonstrates reliable on-site quantification in spiked and real seafood samples.
