Safety and effectiveness of new oral drugs orforglipron and danuglipron for treating type 2 diabetes and obesity

Abstract

In a review of seven randomized trials involving 1037 patients, novel glucagon-like peptide 1 receptor agonists (GLP-1RAs) resulted in a significant reduction in HbA1c levels by 1.03% in patients with type 2 diabetes mellitus (T2DM).

  • GLP-1RAs are associated with a significant weight reduction of 3.26 kg in patients with T2DM compared to controls.
  • In patients with obesity, GLP-1RAs showed a significant weight decrease of 7.52 kg versus controls.
  • The use of GLP-1RAs did not significantly increase the odds of severe hypoglycemia or serious adverse events.
  • There was a noted increase in gastrointestinal adverse events associated with GLP-1RAs, with odds 2.57 times higher than controls.
  • Adverse events leading to treatment discontinuation were also more likely with GLP-1RAs, with odds 2.89 times greater compared to controls.
