Redesigning RNA Structures to Reduce Leakage in CRISPR/Cas12a Systems for More Accurate Genetic Testing

Updated

Abstract

This engineered CRISPR system achieves femtomolar sensitivity, over 10,000-fold higher than a single Cas12a system.

  • A 3'-end DNA extension of scaffold RNA is used to create a conformational switch in the Cas12a system.
  • The design introduces steric hindrance as an activation barrier, allowing for controlled activation instead of self-activation.
  • This framework resolves the conflict between signal enlargement and background variation.
  • The system enables rapid detection of HPV16/18 in under 30 minutes.
