SDS-CRISPR enabled rapid, highly precise single-nucleotide variant detection with attomole sensitivity and detection down to 0.01% variant frequency.

Evidence

This diagnostic platform study combined AlphaFold3-guided CRISPR-Cas12a redesign with a lateral-flow and smartphone readout, then validated performance in 43 glioma tissue samples and plasma cfDNA against immunohistochemistry and next-generation sequencing.

Caveat

The human data mainly show diagnostic concordance in a modest validation set rather than improved clinical outcomes.
