Semaglutide may protect the liver from 5-fluorouracil damage by affecting cell survival signals, mitochondrial cleanup, and inflammation pathways

Updated

Abstract

Semaglutide significantly mitigates 5-fluorouracil-induced liver injury in rats.

  • 5-fluorouracil increased hepatic enzyme levels, oxidative stress, inflammatory markers, and caused histological injury.
  • Pretreatment with semaglutide counteracted the harmful effects of 5-fluorouracil by suppressing oxidative stress and inflammation.
  • Semaglutide enhanced mitophagy through activation of the phosphorylated CREB/PINK1/Parkin pathway.
  • The addition of chloroquine, an autophagy inhibitor, abolished the protective effects of semaglutide.
  • Semaglutide reduced proinflammatory cytokines and oxidative stress markers while enhancing antioxidant activity.
