Semaglutide's heart benefits relate more to the dose taken than to the amount of weight lost

Abstract

Higher maximum semaglutide dose was strongly associated with lower post-landmark risks of all-cause mortality and cardiovascular events.

  • Dose escalation of semaglutide resulted in an additional weight loss of 3.15% per 1 mg increase.
  • Higher doses of semaglutide were linked to reduced risks of all-cause mortality (risk ratio 0.42) and composite cardiovascular events (risk ratio 0.51).
  • Achieved weight loss did not significantly correlate with reduced all-cause mortality or composite cardiovascular events.
  • Semaglutide improved hemoglobin A1c and blood pressure, with both showing strong statistical significance.
  • In matched analyses, semaglutide showed lower cardiovascular event rates compared to other diabetes medications.
  • Transcriptomic analyses indicated high expression of GLP1R in cardiac tissue, particularly in heart cells.
