Semaglutide improves metabolism and thinking ability in a mouse model of Alzheimer’s disease

Abstract

SMGL treatment for 13 weeks in metabolically challenged 5xFAD HFD mice resulted in reduced body weight and improved glucose tolerance.

  • Treatment with SMGL led to normalized cholesterol levels and a restored balance of adiponectin and leptin in 5xFAD HFD mice.
  • Significant reductions in Aβ40 and Aβ42 levels were observed, indicating a potential impact on Alzheimer’s disease pathology.
  • Increased levels of synaptophysin and βIII-tubulin were noted, which are associated with enhanced synaptic function.
  • Immunoreactivity markers for inflammation decreased in the hippocampus and cortex, suggesting anti-inflammatory effects of SMGL.
  • Minimal metabolic responses were observed in WT RD mice, implying that excessive GLP-1 receptor activation may negatively affect neuronal health under normal metabolic conditions.
