Semaglutide and Tirzepatide reduce liver fat absorption and fat breakdown in obese mice fed a high-fat diet

Updated

Abstract

Tirzepatide produced a 24.6% body weight and 30.3% fat mass reduction in diet-induced obese mice after 4 weeks of treatment.

  • Both semaglutide and tirzepatide led to overlapping targeted hepatic proteomic signatures in the liver.
  • Both drugs significantly reduced the levels of CD36 and CPT1A, which are involved in fatty acid import for fat breakdown.
  • No changes were observed in lipogenic enzymes or glucose-handling proteins, indicating a selective modulation of fatty acid metabolism.
  • The findings suggest that GLP-1 receptor activation alone may be sufficient for the main liver response, with GIP primarily enhancing effects outside the liver.
  • The CD36/CPT1A proteomic signature could serve as a benchmark for assessing new GLP-1 and non-GLP-1 therapeutics.
