Different types of aging beta cells separately contribute to pancreatic islet aging and dysfunction

Abstract

Single-cell analysis of the pancreas from 26 donors (ages 20-80) reveals two distinct populations of senescent cells.

  • Senescent cells (SnCs) are identified as having either impaired or retained functional characteristics.
  • One subpopulation of SnCs shows loss of β-cell identity and impaired insulin secretion, along with a proinflammatory response.
  • The other subpopulation retains transcriptional identity and functional competence, exhibiting lower levels of inflammation.
  • Increased immune cell infiltration is associated with the maladaptive SnC subpopulation.
  • These findings highlight the heterogeneity of cellular senescence in the pancreas, which may inform future therapies for type 2 diabetes.
