Gene activity and DNA regulation in individual brain cells of people with PTSD

Abstract

Molecular responses were profiled in over two million nuclei from the dorsolateral prefrontal cortex of 111 human brains with and without PTSD.

  • PTSD is associated with gene alterations in inhibitory neurons, endothelial cells, and microglia.
  • Changes were noted in pathways related to glucocorticoid signaling, GABAergic transmission, and neuroinflammation.
  • Key genes such as SST and FKBP5 showed disruption in cell-type-specific analyses.
  • The study integrates genetic, transcriptomic, and epigenetic data to explore regulatory mechanisms affecting PTSD-related genes.
  • Findings suggest that multiple cell types contribute to the molecular biology of PTSD rather than a single cell type.
