SLC25A28 is a critical weakness in cancers lacking a DNA repair system

Updated

Abstract

Approximately 10% of bladder, upper urinary tract, and endometrial cancers have harmful changes in genes related to nucleotide excision repair.

  • Platinum-based chemotherapy is commonly used for treating tumors lacking functional nucleotide excision repair but is limited by kidney toxicity.
  • A genome-wide CRISPR-Cas9 screen identified the gene SLC25A28 as a key vulnerability in cancer cells lacking the ERCC4 gene.
  • Loss of SLC25A28 was found to significantly reduce cell survival specifically in cancer cells deficient in several nucleotide excision repair genes.
  • Inducible knockout of SLC25A28 in certain cancer models decreased tumor growth in mice.
  • RNA sequencing indicated that loss of SLC25A28 led to increased oxidative stress and DNA damage, suggesting a link to mitochondrial iron management.
