Blocking SLC38A5 May Help Immunotherapy Work Better by Reducing Glutamine Competition Between Brain Tumor Cells and Immune T Cells

Updated

Abstract

The glutamine transporter SLC38A5 may modulate the efficacy of PD-1 immunotherapy in glioblastoma.

  • SLC38A5 is significantly upregulated in human glioblastoma compared to normal brain tissue.
  • Ablation of SLC38A5 sensitized glioblastoma to PD-1 therapy in a manner dependent on CD8+ T cells.
  • SLC38A5 deficiency created a glutamine-enriched tumor microenvironment, enhancing CD8+ T cell function.
  • This deficiency promoted MHC-I-mediated antigen presentation in glioblastoma cells through a glutamine-glutathione-ROS axis.
  • An SLC38A5-targeting nanobody was developed, showing potential to enhance PD-1 therapy and suppress brain tumor growth in vivo.
