SmartPE allows precise gene editing in dicot crop plants

Updated

Abstract

SmartPE achieves 6.3-23.0-fold higher editing efficiencies in Arabidopsis thaliana compared to standard prime editing systems.

  • SmartPE combines a dual-pegRNA strategy with designed mismatched bases and engineered reverse transcriptases.
  • Editing efficiencies in tomato (8.2-38.2-fold) and improvements in soybean and oilseed rape exceed those of conventional prime editing.
  • In tomato hairy roots, SmartPE produced homozygous and heterozygous edits at a significantly higher rate than standard methods.
  • Stable transformations with SmartPE resulted in editing efficiencies of up to 32.3% for seamless genetic modifications.
