Restoring cell recycling to slow eye tissue breakdown by targeting the SNAI1-LAMP3 pathway

Abstract

Sodium iodate (NaIO3) induced lysosomal dysfunction in retinal pigment epithelial cells, impairing autophagy and exacerbating retinal degeneration.

  • NaIO3 triggered lysosomal membrane permeabilization, leading to autophagic flux impairment in RPE cells.
  • RNA sequencing revealed that the gene Lamp3 was downregulated following NaIO3 treatment.
  • Overexpression of LAMP3 alleviated NaIO3-induced lysosomal dysfunction and improved autophagic activity.
  • Increased LAMP3 expression reduced oxidative stress and apoptosis in RPE cells and mitigated retinal degeneration in a mouse model.
  • NaIO3 upregulated the transcription factor SNAI1, which represses LAMP3, and its knockdown enhanced LAMP3 expression.
